First, If you have messed up the slot in the screw I would recommend buying a replacement before getting it out.
The trick is to use the proper size screwdriver that fits the slot in the screw. This usually means you need a 18"+ long screw driver with about a half inch wide bit, minimum of 1mm or .040" thick. You must make sure the screw driver is inline with the axis of the screw before trying to turn it.
Once the screw is messed up, you can make a manual impact screw driver. It takes two people. Clip a vise grip wrench to the screwdriver shank. Have a freind tap on the end of the screwdriver handle with a hammer while you slowly turn the vise grips and screwdriver. The combination of the hammer tapping and the torque of the vise grips will usually break it free (plus the tapping - not hammering - keeps the bit in the slot).
